İkinci sınıfın ilk genetik laboratuvarı dersinden merhaba! Bugün dönem boyunca laboratuvarda çalışacağımız model organizmalardan bahsettik ve ilk model organizma ile başladık: Meyve sineği. Genetik çalışmalar için büyük bir nimet. Bu canlılar için bir besiyer hazırlanıyor. Karbonhidrat ağırlıklı besleniyorlar. Hazırlanmış besiyerler kullandık bu derste ama besiyerin hazırlanışı da oldukça kolay. Bunun için elimde gördüğünüz bu besiyer kaplarına ticari olarak satın alınmış meyve sineği besiyerinden ekleyip üzerine birazcık su koyuyoruz. Çok az bira mayası ekledikten sonra hazırlanmış oluyor. Meyve sinekleri bu besiyere konuluyor ve larvalar olgunlaşmaya başlıyorlar. Erginliğe ne kadar yakın olursa bu canlı, o kadar kahverengi bir görüntü alıyor. Genetik çalışmalar için önemli olmasının sebebi, döllenme süreleri. Çabuk döl elde edebildiğimiz için çaprazlamaları hızlı halledebiliyoruz. Bu derste bu canlıları diseksiyon mikroskobu altında inceledik ve dişi-eril durumlarını kontrol ettik. Ön bacaklarında taraklanmalar (kıl yumağı gibi) olup olmadığına baktık. Eğer varsa erkek yoksa dişi olduğuna kanaat getirdik. Tabii mikroskoba almadan önce bayıltmamız gerekiyordu daha iyi inceleyebilmek için. Uyguladığımız adımlar şunlardı: •Besiyerin ucunda sünger bulunuyordu, öncelikle besiyere hafif hafif vurarak sineklerin aşağı düşmesini sağladık (videoda yaptığım gibi). •Elimizde bir de bayıltma kabı bulunuyordu. Sinekler iyice döküldükten sonra besiyerin süngerini hızla çıkarıp pamuğunu çıkardığımız bayıltma kabına, sinekleri boşalttık. Hepsinin düşmesi için de besiyer kabına vurmaya devam ettik. Bayıltma kabına aldıktan sonra pamukla kapattık ve pamuğun üzerinden 4-5 damla eter damlatıp meyve sineklerinin bayılmalarını bekledik. Yaklaşık 3-4 dakika içerisinde bayıldılar. İncelemeye aldık hemen zira 15 dakika içerisinde kendilerine gelmeye başlıyorlardı. Bizim incelediğimiz tür “white” idi. Gözleri bembeyazdı. See Laungage Is A Funny Thing. It can be viewed as one side of the scale, while numbers make up the other. The original language of numbers...yes....laungage...has its ceiling point. That's per its design. The evolution of numbers gave the term "infinite" life. And through this, became much later "laungage" as you know it today. Without the basic evolution of this number pattern, a circle would have a max size, and there for everything within a maximum number of occurrences. This number can reflect the universe we know with its possible combinations. This maximum, would then give Pi a definite number no longer asking a system to perform an insanity-task. This gives limits and direction to the funnel of time. Regardless if you set a maximum number range or not, each language has it's own maximum combinations as well. Which then when multiplied, gives you the new maximum universal size for the input area requested. Hello lil spiky 👋🏼 🦔🦔🦔 I’ve always liked this image of a cancer cell on the move 😍 You can see at the top of the cell there is a thick band of red (which is the actin cytoskeleton). This is called a lamellipodia structure - it shows the “leading edge” of the cell, ie. the direction in which the cell is moving. ❤️💚💙 There are also lots of spikes at the leading edge of this cell - these are called filopodia. These little spikes are used to sense the cells’ environment - like antennae. You can see there are also some similar structures towards the back of the cell. 💙💚❤️ In the middle is the big fat nucleus in blue. This is where all the cells’ DNA is kept. 💚❤️💙 I’ve had a crazy ass day (including 4 hours in cell culture 😫😂) - hope you’ve all had a good one 😘

Reading weekend I try to read as much as I can. About a year ago I decided that I would like to read a minimum of 10 articles a day. But with an emphasis on quality and reading comprehension, not quantity. It takes quite a long time, but I think it is very important. After a while, it got into my habit and I'm currently increasing the number of papers. Reading a large number of articles allowed me to learn literature very well and to know in depth the field I am currently working on. What is my reading scheme for scientific articles? At the beginning I divide papers into two groups. The first of these are newly published articles. I use the @researcherapp to select them. I follow the best journals and browse all the articles published by them. If any of them seems interesting - I save it and read it in my free time. I enjoy reading articles even from very distant fields from me. I chose very good articles from big journals, e.g. Nature, and read it very carefully. It is very developing and broadening the point of view. I also use Researcher to search new articles in my field using my keywords. Finally, I use Research Gate intensively - I follow many scientists and labs and read their newest papers. When it comes to articles in my field, I start reading them by study a given topic. I browse dozens of articles and choose the most important ones. You can see how my web browser looks like when I select articles 😅 When I read, I focus first on figures and results. If they are interesting and important, I read the entire article. I start by reading Materials and methods to understand researchers. Then I focus on figures and results. At the end I read the discussion and abstract. I usually skip the introduction, in each publication it is very similar. My scheme is very similar to this described by Kath @weekday_neuroscientist If I think that the article is important for my research, relevant to the subject or just interesting, I print it and highlight the most important things. I try to remember Journal, year of publication and lab in which paper was prepared. In the last photo you can see the number of the most important articles for my current project.
